Abstract
U ovom radu provedena je validacija spektroskopskih metoda (metoda za ukupne polifenole (Folin-Ciocalteu metoda), metoda za ukupne flavonoide, metoda za ukupne antocijanine (pH-diferencijalna metoda)) i kromatografske metode za određivanje polifenolnih spojeva. Parametri validacije koji su se određivali bili su: linearnost, granica detekcije, granica kvantifikacije, preciznost, točnost te osjetljivost. Folin Ciocalteu metoda validirana je za galnu kiselinu, (+)-katehin i kvercetin-3-rutinozid, metoda za ukupne flavonoide za (+)-katehin, kvercetin-3-rutinozid i kvercetin, a metoda za ukupne antocijanine za cijanidin-3-galaktozid i cijanidin-3-glukozid. Kromatografska metoda validirana je za sve spomenute polifenole. Utvrđeno je da su spektroskopske metode linearne, s prihvatljivim granicama detekcije i kvantifikacije te s prihvatljivom preciznošću i točnošću. Osjetljivost je bila najbolja za standarde polifenola koji se uobičajeno koriste kao standardi: galna kiselina za ukupne polifenole, (+)-katehin za ukupne flavonoide, cijanidin-3-glukozid za ukupne antocijanine. Kromatografska metoda je također dovoljno precizna i točna, s dobrim granicama detekcije, kvantifikacije, linearnošću te s prihvatljivom osjetljivošću.

Parallel abstract (English)
In this paper, the validation of spectroscopic methods (method for total polyphenols (Folin-Ciocalteu method), method for total flavonoids, method for total anthocyanins (pH differential method)) and chromatographic method for the determination of polyphenol compounds was carried out. Validation parameters which were determined were: linearity, limit of detection, limit of quantification, precision, accuracy and sensitivity. Folin-Ciocalteu method was validated for gallic acid, (+) - catechin and quercetin-3-rutinoside, methods for the total flavonoid content for (+) - catechin, quercetin-3-rutinoside and quercetin and method for total anthocyanins for cyanidin-3- galactoside and cyanidin-3-glucoside. Chromatographic method was validated for all of these polyphenols. It has been shown that spectroscopic methods are linear, with acceptable limits of detection and quantification and with acceptable precision and accuracy. The sensitivity was the best for the polyphenol standards which are commonly used: gallic acid for total polyphenols, (+) - catechin for total flavonoids, cyanidin-3-glucoside for total anthocyanins. Chromatographic method was also sufficiently precise and accurate, with good limits of detection, quantification, linearity and with reasonable sensitivity.
