Abstract
Aronija je voće koje sadrži vrlo velike količine različitih grupa polifenolnih spojeva (antocijanini, fenolne kiseline, flavonoli). Da bi se odredila količina svih prisutnih polifenolnih spojeva, nužno je pronaći odgovarajući ekstrakcijski postupak. Iz tog je razloga u ovom radu ispitan utjecaj različitih uvjeta ekstrakcije (otapala s različitim omjerima vode i metanola, metanol s 0,1 % kloridne kiseline, različito vrijeme ekstrakcije, ekstrakcija ultrazvučnom kupelji ili tresilicom) na ekstraktibilnost polifenola iz aronije. Ukupni polifenoli i ukupni antocijanini određivani su Folin- Ciocalteu i pH-diferencijalnom metodom, a individualni reverzno-faznom tekućinskom kromatografijom visoke djelotvornosti (RP-HPLC). Uz to, razlike u sastavu polifenolnih spojeva u različitim ekstraktima provjerene su i snimanjem njihovih UV/Vis spektara. Kao povoljna metoda za ekstrakciju ukupnih polifenola i ukupnih antocijanina pokazala se 0,1 % metanolna otopina kloridne kiseline uz upotrebu ultrazvuka u trajanju od 15 minuta. Rezultati dobiveni HPLC analizom pokazali su da je za ekstrakciju pojedinih grupa polifenola povoljniji odabir različitih ekstrakcijskih otapala (fenolne kiseline - 50 % metanol u vodi, ultrazvuk, 15 minuta; antocijanini, flavonoli – 0,1 % metanolna otopina kloridne kiseline, ultrazvuk, 15 minuta).

Chokeberry is a fruit that contains high amounts of different groups of polyphenols (anthocyanins, phenolic acids, flavonols). To determine the amount of all polyphenolic compounds present, it is necessary to find a suitable extraction procedure. Therefore, in this study the effects of different extraction conditions (solvents with different ratios of water and methanol and methanol with 0.1% HCl, different extraction times, effect of ultrasound) on the extractability of polyphenols from chokeberry were investigated. Total polyphenols and total anthocyanins were determined by Folin-Ciocalteu and pH differential method and individual by reversed phase high performance liquid chromatography (RP-HPLC). Additionally, differences in the composition of polyphenolic compounds in chokeberry extracts were checked by recording their UV/Vis spectra. 0.1% methanolic hydrochloric acid solution with the use of ultrasound for 15 minutes proved to be a convenient method for the extraction of total polyphenols and anthocyanins from chokeberry. The results obtained by HPLC analysis showed that for the extraction of certain polyphenolic groups it is more favorable to use different solvents (phenolic acid - 50% methanol in water, 15 min ultrasound; anthocyanins and flavonols - 0.1% methanolic hydrochloric acid solution, 15 min ultrasound).
