Abstract
Korištenje lignoceluloznih materijala kao sirovina u biotehnološkoj proizvodnji ovisi o mogudnosti razgradnje lignoceluloznih sastavnica celuloze, hemiceluloze i lignina do fermentabilnih šedera. Filamentozne gljive bijelog truljenja su rijetki organizmi koji imaju sposobnost sinteze ekstracelularnih enzima odgovornih za razgradnju lignoceluloznih sastavnica biljaka. U ovom radu, gljiva Phanerochaete chrysosporium uzgajana je u uvjetima fermentacije na čvrstim nosačima na repinim rezancima (lignocelulozni otpadni materijal) kao supstratu, u laboratorijskim staklenkama pri 27 °C u trajanju od 30 dana. Cilj rada bio je istražiti promjene sastava supstrata tijekom fermentacije s naglaskom na razgradnju lignoceluloznih polimera. U početnom uzorku repinih rezanaca, kao i u uzorcima prikupljenim tijekom fermentacije određeni su udjeli ekstraktivnih tvari, pepela, dušika, celuloze, lignina i pentozana, te gubitak na masi lignoceluloznog materijala po završetku fermentacije. Postignut je gubitak na masi lignoceluloznog materijala (19,62%), te povedanje udjela ekstraktivnih tvari (122%), pepela (41%) i dušika (54,24%) kao posljedica rasta gljive P. chrysosporium. Postignuta je 24%-tna razgradnja lignina (nakon 20 dana fermentacije) i 21%-tna razgradnja pentozana. Nije došlo do razgradnje celuloze pri primjenjenim uvjetima uzgoja.

The use of lignocellulosic materials as a biotechnological production substrate depends mainly on the degradation of lignocellulosic components cellulose, hemicellulose and lignin to fermentable sugars. Filamentous white-rot fungi are among rare microorganisms that produce extracellular enzymes responsible for degradation of plants’ lignocellulosic components.The aim of this study was to cultivate fungus Phanerochaete chrysosporium under solid-state conditions using sugar beet pulp (lignocellulosic waste material) as a substrate and to investiagte the substrate compositional changes during fermentation with emphasis on lignocellulosic polymers degradation. P. chrysosporium was cultivated in laboratory jars at 27 °C for 30 days. The chemical composition of samples collected before and after fermentation was analysed, i.e. substrate weight loss, the contents of total extractives, ash, nitrogen, cellulose, lignin and pentosans. The observed substrate weight loss (19.62%), as well as the increase of total extractives (122%), ash (41%) and nitrogen (54.24%) all indicated the growth of P. chrysosporium. The achieved conversions of lignin (20 days of fermentation) and pentosan were 24% and 21%, respectively. Cellulose degradation was not observed under the applied cultivation conditions.
