Abstract
Cilj ovog završnog rada jest prikazati ovisnost sekundarnog metabolizma plijesni roda Aspergillus o regulaciji Velvet kompleksom. Naime, plijesni roda Aspergillus sekundarnim metabolizmom sintetiziraju mikotoksine. Sekundarni metabolizam plijesni ovog roda reguliran je trimernim proteinskim kompleksom koji se naziva Velvet kompleks. Na ovaj regulator sekundarnog metabolizma od okolišnih faktora najveći utjecaj ima prisutnost svjetla, dok je utjecaj ostalih čimbenika na regulaciju Velvet kompleksom još uvijek predmet istraživanja. Produkcija aflatoksina je pojačana u odsutnosti svjetla što se povezuje s obrambenim mehanizmom plijesni čije je primarno stanište tlo gdje vlada tama i gdje joj prijeti opasnost od kukaca i sličnih organizama, dok je u prisutnosti svjetla produkcija aflatoksina smanjena. Upravo prisutnost svjetla onemogućuje vezanje transkripcijskih faktora VeA i VelB u citoplazmi stanice te transport takvog dimera u jezgru stanice plijesni. U suprotnom, VeA i VelB se povezuju u citoplazmi, transportiraju u jezgru gdje u interakciju stupaju s LeaA što omogućuje pojačanu ekspresiju gena zaduženih za sekundarni metabolizam plijesni, odnosno biosintezu aflatoksina u slučaju plijesni roda Aspergillus kao najpoznatijeg sekundarnog metabolita plijesni ovog roda.
Abstract (english)
The aim of this paper is to present the dependence of the secondary metabolism of Aspergillus on regulation by Velvet complex. The Aspergillus spp. fungi synthesizes mycotoxins by secondary metabolism reactions. Secondary metabolism of these fungi is regulated by trimeric protein complex called Velvet. This secondary metabolism regulator is the most affected by light presence, while the effect of the other environmental factors on this regulator is still under the research. However, aflatoxin production is increased in the light absence, which is related to the defense mechanism of fungi located in the soil where darkness is present and where the risk from insects and similar organisms exists, while in the light presence the aflatoxin production is decreased. The light presence disables the interaction of VeA and VelB transcription factors, and the dimeric complex transport to fungi nucleus. Opposite to that, in the light absence, after interaction in the cytoplasm of the fungal cell, the VeA and VelB are translocated into nucleus and bounded to LeaA which enables increase of the expression of the genes related to secondary metabolism of Aspergillus spp. fungi and aflatoxin biosynthesis as the most famous of all secondary metabolites.
